New preprint: Structural basis for CTCF-mediated chromatin organization

Using in vitro reconstitution and cryo-EM, we show that CTCF dimerization drives higher-order nucleosome assemblies via specific CTCF–CTCF and histone–histone contacts. MicroC and differentiation assays show that disrupting this interface in cells reduces chromatin looping and impairs differentiation.
